The Rome-based outfit were paired with Gregorio Manzano's men in Friday's draw for the last 32 of Europe's second-tier competition.
The 66-year-old also admitted that he was glad to have avoided Manchester United and City, but underlined how difficult it is going to be against the Mattress Makers.
"The draw you have to accept, however, we were lucky to have avoided the English teams," the former Napoli coach told Corriere dello Sport.
"Atletico Madrid are a strong team, and the task is difficult, but we are confident at the Stadio Olimpico.
"We know it's tough, but it's always a pleasure to play against teams of this level," he concluded.
Lazio are currently in fourth in Serie A, two points adrift of leaders Juventus, and host Udinese on Sunday.
